Context
Recently, Indian scientists from IASST and IIT Guwahati developed a new “smart” cancer drug called RK-251. It is designed to become active mainly inside cancer cells by detecting high levels of Reactive Oxygen Species (ROS), thereby reducing damage to healthy cells.
Early laboratory studies showed promising results against triple-negative breast cancer, but further research is needed before human testing.
Mechanism of RK-251
- Targets cancer cells: Cancer cells have high levels of ROS, which switches RK-251 “ON” mainly inside these cells.
- Releases NBDHEX: Once activated, RK-251 releases the anticancer compound NBDHEX.
- Kills cancer cells: NBDHEX blocks GSTP1, a protein that helps cancer cells survive and resist treatment, thereby helping destroy the cancer cells.

Preclinical Findings
- Cancer Model: Showed strong activity against aggressive triple-negative breast cancer cells (MDA-MB-231), with much less effect on healthy cells.
- Toxicity Study: Behavioural study in zebrafish embryos (Danio rerio) showed no obvious signs of toxicity.
- Fluorescence Marker: Drug exhibited the desired fluorescence in the presence of ROS, aiding detection of activation.
Conclusion
RK-251 represents a promising step towards targeted and safer cancer treatment by selectively attacking cancer cells while reducing harm to healthy cells.
Further research and clinical trials will determine its potential as a future alternative to conventional chemotherapy.
